- W2387758464 abstract "Objective:The feasibility of repeated intrathecal administration via lumbar puncture with propofol anesthesia was investigated to provide a new method for repeated intrathecal administration in rats.Methods:24 Sprague-Dawley rats(male,250～300g) were randomly divided into naive group, propofol group,lumbar puncture group and lumbar puncture with saline injection group,6 rats in each group.Rats were anesthetized with propofol(50 mg/kg),then injected transcutaneously at the L5-6 interspace using a 50μl Hamilton syringe.Correct subarachnoid positioning of the tip of the needle was verified by a tail-or paw-flick test.In the experiment,the thermal tail-flick latencies,changes of astrocytes and microglia cells in the spinal cord were determined.Results:No significant difference of thermal tailflick latencies was found between the propofol group,lumbar puncture group,lumbar puncture with saline injection group compared to the naive group(P0.05).There was no significant difference of astrocytes and microglia cells in the spinal cord between experiment rats and the control rats(P0.05). Conclusion:Repeated intrathecal administration via lumbar puncture in rats with propofol anesthesia is safe and effective.This method can be used in the experiments that need repeated intrathecal administration and immediate testing of pain behavior in rats after intrathecal administration" @default.
