- W2388395573 abstract "Objective To investigate the effects of carbon monoxide(CO)inhalation on the mul- tiple organ(lung and intestine)injury induced by lipopolysaccharide(LPS)in rats.Methods Eighteen male Sprague-Dawley rats were randomly divided into three groups.The rats in organ injury group re- ceived LPS 5 mg/kg i.v.,those in control group received normal saline,and those in CO inhalation group received an inhalation of 250 ppm CO after multiple organ injury induced by LPS.The animals were sac- rificed by exsanguinations,and lung and ileum were harvested at 3rd h of observation.The levels of lung tumor necrosis factor-α(TNF-α),interlukin-6(IL-6)and intestine platelet activator factor(PAF),inter- cellular adhesion molecule-1(ICAM-1)were measured by using enzyme-lined immunosorbent assay.The malondialdehyde(MDA)concentration and myeloperoxidase(MPO)activity were detected by using chemical methods.The cell apoptosis was detected by using the flow cytometric method.Pathological changes were observed by histologists.Results LPS challenge resulted in a significant increase in the TNF-α,IL-6,MDA,MPO and apoptotic index of lung by 3.15,2.37-,6.10-,1.36-and 26.50-fold that of control group,respectively,all P<0.05,as well as a significant increase in the PAF,ICAM-1 and apoptotic index of intestine by 2.42-,2.05-,4.43-,2.10-and 5.17-fold that of control group,respective- ly,all P<0.05,accompanied by remarkable organ injury.Either the TNF-α,IL-6,MDA,MPO and apop- totic index of lung or the PAF,ICAM-1,MDA,MPO and apoptotic index of intestinewas significantly de- creased after CO inhalation by 39.5%,44.8%,19.7%,12.1%,81.1%,142.1%,105.6%,343.6%, 110.4 % and 20.5 % as compared with that of organ injury group,respectively,all P<0.05.Moreover, there was a remarkably ameliorated organ injury.Conclusion Carbon monoxide protects multiple organ from injury via anti-oxidation,anti-inflammation and anti-apoptosis,which may play an important role in the pathogenesis of LPS-induced multiple organ injury in rats." @default.
