- W2388552616 abstract "Objective To investigate the effects of PFT-α (p-53 inhibitor, PFT-α), a p53 specific inhibitor, on the apoptosis and p53 target gene (Bax and MDM2) expression in intestinal epithelial cells (IECs) induced by hyperthermic chemotherapy. Methods The primary cultured IECs were divided into 3 groups: control (CON), hyperthermic chemotherapy (HTC), and PFT-α plus hyperthermic chemotherapy (PFT-α plus HTC). IECs were treated with hyperthermic cisplatin (43 ℃, 10 mg/L ) for 30 min, absence or presence of different dosages of PFT-α. The rate of apoptosis was detected by using FCM with Annexin V-FITC and PI double labeling method. The influence of antitumor potency of hyperthermic chemotherapy by PFT-α was analyzed with using methabenzthiazuron (MTT) assay. The expressions of p53, Bax and MDM2 were measured by Western blot. Results FCM analysis showed that apoptosis rate decreased after IECs were exposed to PFT-α in a does-dependent manner. Hyperthermic chemotherapy treatment resulted in the increase of nuclear-to-cytoplasmic p53 ratio. Western blot analysis revealed that PFT-α decreased the nuclear-to-cytoplasmic p53 ratio. The protein levels of Bax and MDM2 were elevated by hyperthermic chemotherapy in IECs, whereas PFT-α attenuated those levels after administration at the doses of 10, 20, 30, and 40 μmol/L. PFT-α at lower dosages did not interfere with the antitumor potency of hyperthermic chemotherapy. Conclusion PFT-α has the potential to protect cancer patients against side effects of anti-cancer hyperthermic chemotherapy. The mechanisms are most likely involved in the changes of p53 nuclear translocation and down-regulation of pro-apoptotic Bax and MDM2. Inhibition of p53 may be the new strategy for attenuating the side effects of hyperthermic chemotherapy." @default.
