- W2388792752 abstract "Objective: To observe the effects of protein kinase B1 (Akt1) and Akt2 gene transfection on invasion and migration of human gastric epithelial cell line GES-1. Methods: The plasmid of p-LXSN-Akt1, which contained the whole Akt1 gene sequence, and p-LXSN-Akt2, which contained the whole Akt2 gene sequence, were used to transfect GES-1 cells with lipofectamine. The GES-1 cells, which were transfected by the empty vector p-LXSN, were the black vector, and the no transfected ones were the control. And then the positive cell clones were selected with antibiotics G418. The protein expressions of Akt1, Akt2 and MMP-2 were measured by Western blotting analysis. Transwell assay was used to analyze the invasion and metastasis capability. The expression of F-actin was detected by immunofluorescence. Results: The GES-1 cell models expressed Akt1 gene (Akt1 group) and Akt2 gene (Akt2 group) were obtained. The protein expressions of MMP-2 were significantly increased in Akt1 and Akt2 groups compared with those of the black vector group and control group (P 0.01). Transwell assay showed that there was a significantly increased cell invasion of GES-1 cells in Akt1 and Akt2 groups (P 0.01). The expression of F-actin was increased correspondingly in Akt1 and Akt2 groups detected by immunofluorescence. Conclusion: Akt1 and Akt2 gene could increase cell invasion and migration, leading to malignant transformation of GES-1 cells." @default.
