- W2389809872 abstract "AIM To establish a method for the determination of oxalate in rat urine by reversed phase high performance liquid chromatography and investigate the relationship of forage and uric oxalate of rat.METHOD To observe the effect of different forage on rats,oxalate was determined by using Agilent SB C_(18)(250 mm×4.6 mm,5 μm)column and Agilent zorbax extend-C_(18)(4.6 mm×12.5 mm,5 μm)guard column.An aqueous solution containing 0.018 mol·L~(-1)potassium dihydrogenphosphate and 0.0025 mol·L~(-1)tetrabutylammonium hydrogensulphate at pH 2.5～2.7 was used as mobile phase.Flow rate was 1.5 ml·min~(-1) at 25 ℃.Injection volume was 20 μl.and detected at 210 nm.Deproteinization of urine samples was carried out by well mixing 1.0 ml urine sample with sulfosalicylic acid at 4 ℃ for 30 min.RESULT The detection limit was 0.4 μg·ml~(-1).The linear range was 62.50～4000.00 μg·ml~(-1).The average recovery was 103.49%,RSD of intra-day and inter-day were less than 11.56%.When rats were served different forage,their uric oxalate content fluctuated obviously.CONCLUSION The method is simple,sensitive and producible for the research of formation of uric calcium oxalate stone." @default.
