- W2390000102 abstract "【Objective】This research studied the effects of duck-origin NDV infection on immune function of SPF ducks.【Method】120SPF ducks(pre-vaccinated with inactivated H5N1inactivated oil-emulsion vaccine)were randomly divided into three groups,40in each group,intravenous infection with duckorigin NDV group,intraocular-nasal infection with duck-origin NDV group and control group.Ducks in infection groups were inoculated with 106 ELD50 /0.2mL duck-origin NDV,while the control ducks were inoculated with normal saline.The morbidity and mortality of each group were observed.Ducks from each group were randomly killed at different times.Spleens and bursas were collected and paraffin sections were made before the cell apoptosis was detected using TUNEL.In addition,blood samples were randomly collected from ducks in each group to measure and evaluate blood biochemical value,blood routine examination,lymphocyte transformation rate,effect of H5N1inactivated oil-emulsion vaccine on antibody,and CD4+/CD8+ratio in peripheral blood.【Result】Most of the ducks infected experienced dyspnea and hemorrhage in lung and proventriculus,and the morbidity and mortality were 100% and 32.5%,respectively. Blood routine examination showed RBC and HGB had a significant(P0.05)or remarked significant(P0.01)increase followed by a decrease compare to the control group.PLT and WBC in intravenous infection with duck-origin NDV group and intraocular-nasal infection with duck-origin NDV group significantly decreased compared with control group.Blood biochemical values showed activities of ALT,AST, LDH,CK,and AMY in intravenous infection with duck-origin NDV group and intraocular-nasal infection with duck-origin NDV group had a significant(P0.05)or remarked significant(P0.01)increase.TP level significantly(P0.05)or remarked significantly(P0.01)decreased compared with control group. HI titers post H5N1vaccination showed no significant differences compared with control group.Lymphocyte transformation rates showed intravenous infection with duck-origin NDV group and intraocular-nasal infection with duck-origin NDV group had a significantly(P0.05)or remarked significantly(P0.01) lower level than control group.Flow cytometric analysis showed intravenous infection with duck-origin NDV group and intraocular-nasal infection had a similar change in CD4+/CD8+ratio with a decrease to normal level following an increase.Lymphocytes apoptosis presented in spleens and bursas especially in aged ducks.【Conclusion】SPF ducks were immunosuppressed after infection with duck-origin NDV." @default.
