- W2390467085 abstract "Purpose To compare the difference of serum leptin levels among the normal weight, overweight and obese subjects,and to observe the influence of metformin therapy on body weight and leptin level in obese and overweight subjects. Methods Twenty-eight persons with normal BMI were detected the serum leptin levels as control normal range. Sixty cases with overweight or obesity diagnosed by BMI25 or BMI30 and excluded polycystic ovary syndrome (PCOS) and diabetes mellitus were used as treament group. One month after low-calorie diet, 60 patients were administrated 500 mg metformin orally 3 times daily for 5 months,and their body weight, waist-hip ratio, blood pressure, the fasting and post-prandial glucose and insulin levels,plasma lipid profile,and serum leptin concentrations were measured at baseline time, before and after metformoin therapy. The levels of leptin was detected with RIA used the kit bought from Linco Company in U. S. A. Results The normal range of serum leptin was (5. 65±2. 53)ng/mL and was significantly higher in female than that in male. Fifty-two of 60 patients completed the study, whose lep-tinlevel[(23.51±8. 08)ng/mL] before clean-up was significantly higher than normal group. Eight of 60 had to withdraw due to the digestive side effect. However, there was no significant difference between male and female in 52 cases. Among these 52 patients, the leptin concentrations in 34 obese patients were higher than that in 18 overweight ones(P = 0.009 4). The levels of serum leptin decreased slightly after one-month low-calorie diet, indicating no significance. After 5 months of metformin treatment, BMI, WHR, serum lep-tin,fasting and post-prandial insulin,as well as TG and TC decreased markedly(P0.01) , but the HDL-C level increased obviously( P0.01). Increased concentration of serum leptin before treament correlated positively with the degree of weight, waistline, fasting and post-prandial insulin( all P0.05), and passively with FT4 level. Similarly did the reduction of leptin, and it related negatively to HDL. Conclusions The serum leptin level increased in overweight and obese patients. Metformin therapy reduced BMI and WHR in these subjects, and its mechanism may be related with the reduction of leptin levels and insulin-resistance." @default.
