- W2390933532 abstract "Objective To observe the antagonism of selenium to arsenic-induced adverse effect in the activity of SOD and GSH-Px,the content of MDA,and the expression of ICAM-1 and VCAM-1 in human umbilical vein endothelial cells(HUECV). Methods The density of HUVEC in logarithmic growth phase was adjusted to 1.0 ×105/ml,then the cells were divided into two groups and were treated as follows, one group was treated with arsenic solution at different doses(0,0.05,0.1,0.5,1.5,3,6 and 12 μmol/L),the other group was treated with the same doses of arsenic plus sodium selenite(0.1 μmol/L). The medium were collected after 48 h-culture to determine the activity of superoxide dismutase(SOD) and glutathione peroxidase(GSH-Px) and the content of malondialdehyde(MDA) by spectrophotometry. The expression of intercellular adhesion molecule-1(ICAM-1) and vascular cell adhesion molecule-1(VCAM-1) were detected by enzymelinked immunosorbent assay(ELISA). Results With the increase of arsenic exposure,the arsenic group showed a gradual decline trend in the activity of SOD,GSH-Px,while an upward trend in the content of MDA and the expression of ICAM-1 and VCAM-1. Selenium showed a significant antagonism to lower level arsenic-induced adverse effect in the activity of SOD and GSH-Px,the content of MDA,and the expression of ICAM-1 and VCAM-1 in human umbilical vein endothelial cells.Conclusion Exposure to arsenic in a certain level can cause decrease of antioxidation of vascular endothelial cell(VEC) and increase of lipid peroxidation and up-regulated expression of ICAM-1 and VCAM-1. Selenium supplying may have antagonism to the adverse effect of arsenic." @default.
