- W2391160629 abstract "Objective: To study the effect of carvedilol on apoptosis of cardiomyocyte and the expression of apoptosis-related bcl and bax genes induced by ischemia-reperfusion in rats. Methods: 30 Wistar rats were divided randomly into three equal groups. In animals of the sham-operation group, the heart was exposed but the coronary artery was not ligated; in rats of the ischemia-reperfusion (I/R) group, the left anterior descending coronary artery was ligated for 30 minutes and untied thereafter; rats of the treatment group were subjected to the same operation as in animals of the I/R group. The rats in the treatment group were given each 20 mg·kg~(-1) (1 mL in volume) of carvedilol administred by gastrogavage 2 h prior to, immediately after and every 24 h following the operation while those in the other 2 groups were given each 0.9% sodium chloride solution of the same amount, in the same manner and at the same time points as in rats of the treatment group. 48 h after the operation, the animals were sacrificed and the number of apoptotic cardiomyocytes in the heart were determined with the Tannel method and the expression of bcl-2 and bax genes was assayed with the immunohistochemical method and in situ hybridization. Results: The numbers of apoptotic cardiomyocytes were (36.8±9.0), (0.0±0.0) and (9.5±3.0) per visual field of high power objective in the I/R group, sham operation group and treatment group, respectively, the differences being highly significant among the three groups (P0.01). The bel-2/bax protein ratio of the three groups in the same order were (1.41±0.21), (1.52±0.20) and (2.20±0.24), respectively. The difference between the treatment group and I/R group was significant(P0.05). Conclusion: Carvedilol was shown to exert a definite inhibitory effect on the apoptosis of cardiomyocyte induced by ischemia-reperfusion possibly by increasing the bcl-2/bax protein ratio in the cardiomyocytes." @default.
