- W2391590026 abstract "Objective To observe the skeletal malformations in fetal rats treated with retinoic acid and study the pathogenic mechanism of skeletal malformations.Methods Time-mated pregnant SD rats were fed either 125 mg/kg of all-trans-retinoic acid or vehicle on gestational day 10,and the fetuses were removed near term.Twenty fetuses from experimental group and 10 from control group were ran- domly selected.Skeletal anatomy was studied after alcian blue and alizarin red staining.Results Body weight of experimental group was much less than that of control group(P<0.05).All control fetuses were normal,whereas 18 of 20 experimental fetuses had skeletal malformations.The most common mal- formations were long bone malformations(80%),absence of carpal or metacarpal or tarsal or metatarsal or digits(80%),malformation of compages thoracis(80%),vertebrae malformations(75%)and spinal bifida(65%).Conclusion Retinoic acid has a strong and multi-regional teratogenic effect on the skele- ton of fetal rat.The rat model of retinoic acid is a useful tool for investigation of congenital skeletal mal- formations." @default.
