- W2391857140 abstract "Objective To study the relationship between liver dysfunction and growth hormone(GH)resistance in rats after cardiopulmonary bypass(CPB).Methods The CPB model in rats was established.The rats were randomly divided into GH pretreatment group(GH group)and CPB group.The serum levels of GH,GH binding protein(GHBP),endotoxin,tumor necrosis factor(TNF)-α,liver function and prealbumin were detected respectively at different intervals after CPB.Simultaneously,the liver tissues were collected for the detection of the GH receptor(GHR)mRNA by RT-PCR analysis.Results Compared with pre-CPB,the serum levels of LPS,TNF-αwere significantly increased and the lesion of liver function was apparent in both groups.Meanwhile,the expression of GHR mRNA in liver tissues was also decreased obviously in both groups.However,The serum levels of LPS and TNF-αat the same time point in GH group were significantly lower than those in CPB group(P0.05 or P0.01).The serum level of GHBP and the expression of GHR mRNA in liver tissues at the same time point in GH group were significantly higher than those in CPB group(P0.05 or P0.01).Compared with CPB group,the liver function at the same time point improved transparently in GH group.The serum level of GHBP and the expression of GHR mRNA had a significant negative correlation with the levels of bilirubin,LPS and TNF-α(P0.01),and a positive correlation with prealbumin(P0.01).Conclusion There are obvious liver injury and downregulation of GH/GHR axis during early stage of CPB because of the GHR mRNA expression inhibition resulting from inflammatory response.It may be one of the important factors causing the pathophysiologic changes of liver during CPB.GH pretreatment could relieve inflammatory response,up-regulate the axis of GH/GHR,and improve the liver function." @default.
