- W2392080509 abstract "ObjectiveTo investigate the effect of the adoptive transfer of T cells and B cells from the normal pregnancy mice on pregnant outcome of the abortion-prone mice. MethodsThe experiments were performed in following two groups: using ♀CBA/J×♂BALB/c matings as the normal pregnancy model and ♀CBA/J×♂DBA/2 matings as the abortion-prone model. We tested the proliferative capacity and IL-2 production by pregnant CBA/J splenocytes in response to paternal splenocytes as stimulator cells in an one-way mixed lymphocyte reaction(MLR) and counted the embryo resorption rate at day 14 of gestation. Thereafter,purified T and B cells were obtained from spleens of the pregnant CBA/J mice using magnetic beads at day 9 of gestation,and then injected intravenously into the pregnant CBA/J mice mated with DBA/2 males at day 4 of gestation (time of implantation). The embryo resorption rate of recipient mice was counted at day 14 of gestation to evaluate the effect of adoptive transfer of T cells from the normal pregnancy mice on pregnant outcome of the abortion-prone mice. ResultsThe embryo resorption rate of the ♀CBA/J×♂BALB/c matings decreased significantly,and in vitro the proliferative index and IL-2 production by pregnant CBA/J splenocytes in response to paternal splenocytes also decreased significantly as compared to that of the ♀CBA/J×♂DBA/2 matings. In the recipient CBA/J mice,these adoptively transferred T cells from the normal pregnancy mice can increase fetal viability,whereas the adoptive transfer of T cells from he abortion-prone mice or B cells from the normal pregnancy mice did not reduce the embryo resorption rate. ConclusionThe adoptive transfer of T cells from the normal pregnancy mice at the early stage of gestation can induce maternal-fetal immuno-tolerance and prevent maternal rejection to the allogeneic fetus,thus leading to the embryo resorption rate of the abortion-prone model to that in normal pregnancy model." @default.
