- W2393621154 abstract "Goats were randomly allotted to 4 groups of 7,7,6 and 6 animals, respectively.Estrus was synchronized with an 1.5 mg progestagen ear implant left in place for 11 days and two injections of each 5 mg of PGF_ 2α administered i.m. at a 12 h interval on the day of implant removal.Beginning two days before implant removal, does of Group 1,2 and 3 received s.c. injections of 4,4,2,2,2 and 2 AU pFSH at 12 intervals. In Group 4,double doses of pFSH were administered at 24 h intervals. In Group 1,pFSH was pure, while in Groups 2,3 and 4,pFSH was supplemented with 40% pLH.Animals of Groups 1 and 3 received two additional injections of 200 μg pLH together with the last two pFSH-injections.Prior to implant removal goats were fitted with permanently indwelling jugular catheters for repeated blood collection.Concentration of estradiol-17β(E_2) and follicle stimulating hormone(FSH) were measured by means of enzyme-linked immunosorbent assay(ELISA).After removal of the ear implant animals were checked for estrus at 8 h intervals with the adid of an aproned buck.Goats showing signs of estrus were mated a fertile buck once or twice daily as long as they were receptive. The ovarian structures were oberserved by laparoscopy one day before embryo collection. On day 6 or 7 after the last mating, embryos were collected transcervically. The additional pLH-injections at the end of the pFSH-treatment shortened the intervals from implant removal to the onset of the E_2-surge,time of E_2-maximum and end of the E_2-surge.It was found that the E_2-courses were highly varible. The interval from implant removal to the maximum of the E_2-concentration was relatively constant, varying from 28.7 to 38.9 h.The various superovulatory treatment regimes showed no significant effects on the secretion of FSH or on ovarian response and embryo yield.Animals treated with pFSH/40% pLH without an additional pLH injection had more corpora lutea, more embryos and ova and more transferable embryos per doe as compared to the other treatment groups.Significant relationships were found between the number of corpora lutea on the one hand and E_2-maximum(r=0.44,P0.05) and duration of E_2-surge(r=-0.52,P0.01) on the other hand.Significant correlations were also observed between the onset of the second FSH-surge or the time of the second FSH-maximum and the number of corpora lutea(r=0.54,P0.05 and r=0.52,P0.05,respectively).It is concluded that superovulation with pFSH supplemented with 40% pLH leads to satisfactory ovulation rates and number of transferable embryos. The maximum of the E_2-concentration which persistently occurs between 29 and 39 h after implant removal, may be used to distinguish between potentially good and poor embryo donors." @default.
