• W2393811226 abstract "[Objective]To compare the level of NOSmRNA expression and protein expression in the primary organs of rat under different exercise loads,to study the effect of NO at gene level on body in movement.[Methods]By the technique of reverse transcriptase polymerase chain reaction(RT-PCR)to test the expression of NOS subtype(eNOS,nNOS,iNOS),by the mean of Western blot to test the expression of protein expression in different body tissues.[Results]We found that there were some mRNA expression in the tissues of cardiac muscle,brain and vastus lateralis.The level of mRNA expression of eNOS,nNOS,iNOS in the moderate intensity group was obviously higher than the control group.The level of mRNA expression of iNOS in vastus lateralis in the fatigue group was obviously lower than the moderate intensity group.There was no significant difference between different exercise loads on the levels of mRNA expression of both eNOS and nNOS in the tissue of cardiac muscle.The levels of mRNA expression of iNOS in the moderate intensity group and fatigue group were obviously higher than the control group(P﹤0.01).The level of mRNA expression of both eNOS and nNOS were obviously higher after moderate intensity exercise in tissue of brain(P﹤0.05).The level of mRNA expression of nNOS after fatigue was obviously lower than that of the moderate intensity exercise in tissue of brain(P﹤0.05).By the mean of Western blot,we found that the level of protein expression of eNOS,nNOS,iNOS in the moderate intensity group were obviously higher than the control group and fatigue group,and the level of protein expression in the tissue of vastus lateralis in the fatigue group was obviously lower.The level of protein expression of both eNOS and iNOS in the moderate intensity group were obviously higher than the control group,and the level of protein expression of nNOS in the tissue of brain in the fatigue group was the least.After moderate intensity exercise and fatigue,the level of protein expression of iNOS were getting higher,and after fatigue that was the highest.[Conclusion]The moderate intensity exercise is beneficial to both the NOSmRNA expression and protein expression;fatigue exercise is unfavorable to the mRNA expression of nNOS,eNOS and protein expression." @default.
