- W2393987344 abstract "OBJECTIVE To establish a HPLC-UV assay for the determination of norelgestromin(NGMN, the major active metabolite of norgestimate) in serum samples and to study its pharmacokinetics in rats. METHODS NGMN was analyzed on Spherisob C_ 18 column with methanol-water-acetic ether(5∶4∶2) mobile phase at 1.0 mL·min~ -1 flow rate. The wave-length of UV detector was set at 254 nm. A single dose of 0.3 mg·kg~ -1 norgestimate was ig in rats. The concentrations of NGMN in serum at different time were assayed by HPLC-UV method. The pharmacokinetic parameters of NGMN were calculated by program 3P87.RESULTS NGMN is the major metabolite of norgestimate in rats. The calibration curve was linear in the range of 10～50 μg·L~ -1 (r=0.999 6) and the quantification limit was 2 μg·L~ -1 . The recovery of NGMN was between 89.6% and 90.1%, and the within-day and between-day RSD were less than 5%. The concentration-time curve ofNGMN was fitted to a two-compartment model with first order absorption. The pharmacokinetic parameters of NGMN were(2.77±0.14)h for t_ 1/2α ,(31.07±1.42)h for t_ 1/2β ,(2.54±0.65)h for t_ max ,(24.8±5.56)μg·L~ -1 for ρ_ max ,(98.08±6.25)μg·h·L~ -1 for AUC.CONCLUSION This method is simple, accurate and steady, and it can be used to study the pharmacokinetics of NGMN." @default.
