- W2394021437 abstract "Objective To investigate the effect of lidocaine on the expression of Bax protein and cell apoptosis in rat brain after ischemic injury,amassing experimental data for drug choice in clinics.Methods Rat model of cerebral embolism was developed.Forty male SD rats were divided into four groups at random,10 rats each.Group Ⅰ served as blank control,group Ⅱ was sham-operation control group,group Ⅲ underwent operation,and group Ⅳ was treated with lidocanine.The rats were sacrificed by cervical luxation at 24 h after operation.The rat brain was taken out of the skull and placeol on ice plate at once.Then left hippocampus was separated and homogenized,and the supernatant was taken,to detect the level of glutamate by high performance liquid chromatography.The right hippocampus was isolated,fixed with polyformaldehyde,sectioned,and stained with HE to check the structure of tissue.The level of Bax protein was assayed by immunochemistry.TUNEL was employed to determine the cell apoptosis.Results For the results of TUNNEL,(1)there was much less apoptosis in control group,and less apoptosis in sham-operation group,the difference between these two groups being not significant;(2)the rate of cell apoptosis in operated group increased significantly,compared with control group(P0.05);(3)the rate of cell apoptosis in lidocaine-treated group decreased significantly,compared with control group.Immunochemical findings:the Bax protein was expressed in each group;(1)the number of Bax-positive cells in the sham-operated control group was more than that in the control group,but there was no significant difference(P0.05);(2) the number of positive cells in lidocaine-treated group differed(P0.05)less remarkably than that in the operated group,and difference between these two groups was significant(P0.05).Conclusions Lidocaine reduces the expression of Bax in nerve cells.Lidocaine possibly inhibits the apoptosis of nerve cells." @default.
