- W2394339440 abstract "Objective: To explore a practical method of separation and culture in vitro of mesenchymal stem cells (MSCs) from bone marrow of the adult rats, and to offer reference to its actual application. Methods: MSCs from rat bone marrow were separated and purifiedby means of gradient centrifugation and adherence to the culture plastic, then the cells were expanded by subculturing successively and Alkaline Phosphatase (AKP) of the separated cells was tested. Results: MSCs, separated from the bone marrow of the adult rats, were collected in higher purity by means of gradient centrifugation and still kept the cells' activity. These cells, cultured in vitro, grew in an adherent mode on the wall and had a ball-like appearance. They were able to greatly proliferate after 3～4 days of culture and from the cell clones having the analogic cell shape . Alkaline phospha tase(AKP) stain of the separated cells showed strong positive results.Conclusion: It is a simple and practical method to separate MSCs from the bone marrow of adult rats by means of gradient centrifugationand adherence to the culture plastic. MSCs can greatly proliferate after ashort period of culture in vitro. Higher purity of MSCs,separated and cultured in vitro, will be very useful for its further research and applicatin." @default.
