- W2394365502 abstract "Objective To evaluate the effect of recombinant adenovirus vector mediated heme oxygenase-1 gene transfer on discordant cardiac xenografts.Methods Stable cervical cardiac xenotransplantation model of guinea pigs to SD rats was successfully established by Heron's cuff vessel anastomosis technique combining of the use of purified Chinese cobra venom factor.A total of 24 guinea pigs and 24 SD rats were divided randomly into three groups: groupⅠ(control group),group Ⅱ(adenoviral blank-vector group) and group Ⅲ(gene transfer group),The donor heart's coronary arteries were perfused ex vivo with HTK solution containing viral particle of Ad.HO-1 in group Ⅲ,In group Ⅱ the donor heart were perfused with HTK solution containing viral particle of adenoviral blank-vector and virus-free HTK solution in the groupⅠ.The survival time was recorded,the xenografts were collected after cardiac arrested.Gene product expression in tissue was quantified by immunoassay,reverse transcription-polymerase chain reaction and visualized or localized by immunohistochemical staining.Results RT-PCR demonstrated the transcription of the exogenous HO-1 gene in the xenograft.Immunohistochemical examination confirmed that HO-1 protein was successfully expressed in perivascular myocardial cell and coronary capillaries.The group of HO-1 gene transfer has less inflammatory cell infiltration in the xenograft.Expression of exogenous gene attenuated macrophage and nature killer cell infiltration and decreased myocardial cell apoptosis and significantly prolonged average xenograft survival timen=8,(40±3)h],compared with in adenoviral blank-vectorn=8,(21±2)h,P0.01] and in control n=8,(20±3)h,P0.01].Conclusions During the donor heart ex vivo cold ischemia time,intracoronary perfusion of recombinant adenovirus vector mediated heme oxygenase-1 gene demonstrated successful transcription in xenografts,the expression of HO-1 gene can relieve impairment of immune reaction and significantly prolonged xenograft survival time.This study on gene transfer of the HO-1 gene may be the prospect of the clinical xenotransplantation." @default.
