The main difficulty facing the users of these methods is the lack of access to the specialized equipment needed to obtain the best results. It is necessary to use a machine that freezes samples while under pressures of up to 30,000 lb per square inch. Freeze-substitution (FS) is the process whereby the cell water is replaced, or “substituted,” with an organic solvent. It serves the same function as dehydration at room temperature—that is, replacing the cell water with a substance that is miscible with resins. However, FS must be done below a temperature of –70°C so the frozen water in the cell doesn't “recrystallize” into ice. There are a number of different solvents that can be used for FS; however, acetone and methanol are the most popular. The chapter focuses on embryos. However, the method can also be applied to dissected adult tissues, such as ovaries and testes." @default.
