- W2400407712 abstract "Abstract Despite decades of research, successful treatment of affective disorders has remained a significant problem. While conventional pharmacotherapy may offer relief to some, a sizable proportion of patients show inadequate response. Interest in the antidepressant effects of ketamine, an antagonist at N-methyl- d -aspartate receptors throughout the brain, has opened a new venue. Valued for its anesthetic properties in surgery, it has been shown that a single subanesthetic dosage can alleviate both unipolar and bipolar depressive symptoms, with relatively prolonged duration and rapid onset of action. At the same time, knowledge about long-term effects and overall patient safety is restricted and concerns have been raised about potential for misuse. In addition, previous studies have largely failed to address crucial methodological requirements. However, as this chapter will outline, ketamine may provide a scaffold for future antidepressant drugs and help elucidate mechanisms of more efficacious pharmacological interventions." @default.
