- W2404735529 abstract "Transmitter-gated ion channels are multisubunit membrane-spanning receptors that serve as rapid signal transduction devices regulating the flow of cations or anions through the cell membrane. Cell type-specific flexibility in neurotransmission is accomplished by a mUltiplicity of channel variants based on the combinatorial assembly of structurally related subunits. The heteromeric subunit structure also permits synaptic plasticity by changes in subunit composition and subunit-specific posttranslational modification. In addition, receptor heterogeneity offers the potential for cell type-specific drug targeting. By gating the flow of chloride ions, GABAA receptors mediate the major inhibitory neurotransmission in the central nervous system (CNS) and, as apparent from their drug induced modulation, contribute to the regulation of anxiety, vigilance, memory, epilep togenic activity, and muscle tension. Based on a repertoire of subunits encoded by at least 15 genes (al-6, [31-3, ')'1-3, 8, pl-2, with splice variants and homologues in nonmam malian species) (Barnard et al., 1992; Wisden and Seeburg, 1992; ffrench-Constant et aI., 1993; Harvey et al., 1993; Macdonald and Olsen, 1994; Mohler et aI., 1995), GABAA receptors are prime examples for receptor heterogeneity in the CNS. By investigating the physiological and pharmacological significance of GABAA receptor SUbtypes, in combi nation with their molecular architecture and cellular localization, it will be possible to identify the neuronal circuits regulating the above-mentioned CNS states and to provide ~" @default.
