- W2414275791 abstract "Human monocytes show a rapid and transient increase of free cytosolic Ca2+ ([Ca2+]i) after exposure to recombinant MCP-1 (rMCP-1; Sozzani et al. 1991. J. Immunol. 147: 2215). Influx across plasma membrane receptor operated Ca2+ channels (ROCs) appear to be the unique mechanism responsible for this effect. This conclusion is supported by several observations: i) the rise in [Ca2+]i is dependent on the presence of extracellular Ca2+; ii) the rise in [Ca2+]i and Ca2+ influx, measured by Mn2+ Fura-2 fluorescence quenching, are completely blocked by Ni2+ and by SC38249, two Ca2+ channel inhibitors; iii) rMCP-1 and MG-63-derived MCP-1 do not induce IP3 formation nor inositol lipids turnover. Chemotactic response to rMCP-1, Rantes and MIP-la could be inhibited by C-I, a serine/threonine kinase inhibitor, and by Erbstatin and Genistein, two tyrosine kinase inhibitors. However, the three proteins showed a different degree of sensitivity with MIP-lα>Rantes>rMCP-1. On the contrary, neutrophil migration to IL-8 (a member of the Chemokine-α subfamily) was inhibited by tyrosine kinase inhibitors but increased by C-I. Finally, we observed that rMCP-1, Rantes and MIP-la induced both homologous and heterologous desensitization assessed by Fura-2 fluorescence. rMCP-1 desensitized for Rantes and MIP-lα, and Rantes and MIP-lα desensitized each one for the other. However, both Rantes and MIP-1α were ineffective with respect to rMCP-1.
