- W2415236774 abstract "Genetic studies on the gram-positive bacterium Bacillus subtilis have reached a level of sophistication second only to those on E. coli. Much progress has been made in the development of generalized cloning systems. Those based on plasmids suffer from a problem of instability that has not yet been overcome. The reason for this instability may be related to the scarcity of naturally occurring plasmids in B. subtilis. Most of the vectors in use were derived from other gram-positive bacteria and, therefore, may not be well adapted to their new host. In contrast, temperate phages, which are relatively common in B. subtilis strains, appear to provide a much more stable and reliable basis for cloning. This chapter discusses a few naturally occurring plasmids that have been found in strains of B. subtilis. Most of the useful vectors has been originally isolated from Staphylococcus aureus and were subsequently shown to be capable of stable autonomous replication in B. subtilis. The chapter also discusses the salient properties of three of the most extensively used plasmids, pUB110, pC194, and pE194, and of the other plasmids." @default.
