- W2415442654 abstract "The three-dimensional organization of the plasmalemmal vesicles in the peripheral zone of endothelial cells was analyzed in three series of sections of frog mesenteric capillaries. The series contained 21, 21, and 22 serial sections, respectively. The thickness of the single sections was about 150 Å, permitting each vesicle to be contained in an average of 6–7 consecutive sections. The spatial connections between vesicles and the cell membrane were ascertained by following the continuity of the unit membranes. Although about 70% of the vesicles appear “free” in individual sections, following the vesicular membrane in adjacent sections revealed connections to other vesicles and/or to the cell surface. Thus, among 427 vesicles contained in the series, only 3 vesicles were possibly free and isolated, since the connection to other vesicles or the cell membrane was not distinct. If free, they represent less than 1% of the plasmalemmal vesicles in the examined volume of the endothelial cells. No direct transendothelial channels were found in the three series, but in a separate series of thin sections a communication between the blood and the abluminal open part of an intercellular cleft was found. The study strongly supports the recent proposal (M. Bundgaard, J. Frøkj˦r-Jensen, and C. Crone, 1979, Proc. Nat. Acad. Sci. USA 76, 6439–6442) that endothelial vesicles are part of cluster-like invaginations from the cell surfaces and not freely moving entities, as generally believed. It is proposed that the transendothelial channels formed by fusion of the innermost vesicles in the invaginations from the two sides of the capillary wall represent the “large” pores in noninflamed continuous capillaries." @default.
