- W2415672671 abstract "A new technique for the mechanical separation of the two major smooth muscle layers of the human oviductal isthmus was utilized for in vitro studies of contractility. This approach opened the possibility for elucidating a differentiated contractile response to the “natural” prostaglandnis (PGs) (E1, E2, and F2α). Small strips from the respective muscle layers were mounted in organ chambers at 37 ° C for isometric contractility recording. The addition of PGE1 or PGE2 caused a concentration-dependent inhibition of the spontaneous phasic activity of the circular muscle preparations. The longitudinal preparations exhibited a biphasic response to PGE1 and PGE2 with inhibited contractility at low concentrations and a marked stimulation at high concentrations. PGF2α evoked a strong, monophasic, stimulatory response in both layers, characterized in longitudinal specimens mainly by an elevated tone and in circular specimens by an increased frequency of contractions. Differentiated contractile responses of the two muscle layers in the human oviductal isthmus to prostaglandins E and F were thus clearly proven. It is suggested that the demonstrated different characteristics of these two smooth muscle systems are essential for normal tubal function." @default.
