- W2419167137 abstract "Abstract Fifty serum samples from 35 jaundiced patients were studied with reverse phase partition chromatography of bilirubin Pigments I and II and an organic solvent partition of their diazotized sulfanilic acid derivatives. The 2 methods did not correlate well in our experience. More than 50 per cent of patients in each etiological group were shown by both methods to have more Pigment I than II. Serial determinations of Pigments I and II in patients dying of liver failure failed to reveal any change in their distribution. These patients were able to maintain significant serum levels of Pigment II as well as Pigment I right up to the time of death. It has been concluded that fractionation of the individual bilirubin pigments is neither of diagnostic nor prognostic value. Also it has been determined that the alkali-stable, direct reacting bilirubin is not important diagnostically. An alkali-stable, direct reacting fraction was found for both Pigments I and II. The ability to separate conjugated from free bilirubin was utilised to judge the optimal time for reading the direct diazo reaction. The ratio of conjugated to conjugated plus free bilirubin as quantitated from a partition column was compared with the one minute/total, 5 minute/total, and 15 minute/total diazo reactions, respectively. The best correlation was seen to be with the one minute/total value. This supports the routine determination of the direct reaction at one minute. It was observed that the diazo time reaction curve for Pigments I and II were similar up to 30 minutes. Upon the addition of methanol to Pigment I, a small additional color development was observed." @default.
