- W2462102845 abstract "Whole body oxygen uptake (VO2) is an indicator of aerobic fitness, but there is a need to better understand how local factors, such as muscle oxygenation, relates to running performance. PURPOSE: To describe the relationship between oxygen uptake and near infrared spectroscopy (NIRS) parameters of muscle oxygenation during treadmill running. METHODS: Five trained runners participated in this study. A NIRS unit was secured over the vastus lateralis and tissue saturation index (TSI), oxygenated hemoglobin (O2Hb), and deoxygenated hemoglobin (HHb), were recorded at 10Hz. Breath-by-breath VO2 was measured with a metabolic cart. Each runner performed an incremental speed running protocol, where he/she ran at a given speed for four minutes, followed by four minutes of recovery, before completing a VO2max test. One speed, approximating self-reported easy training pace, was performed twice to allow for reliability analysis. Before each running trial, subjects walked at 3mph for one minute to serve as a reference measurment. Walking data were subtracted from steady state running data to compute a relative measurement for each NIRS parameter. Data from each variable were then normalized to the maximum measurement achieved during the VO2max test. Intrasession reliability for VO2 and NIRS parameters were analyzed by computing intraclass correlation coefficients (ICC2,1) from the two trials performed at identical speeds. Linear mixed effects models were used to analyze the relationship between each NIRS parameter across multiple physiologic intensities achieved, with significance set at p<0.05 a priori. RESULTS: ICC2,1 for VO2, TSI, O2Hb, and HHB were excellent (>0.90 for all). Mean ± SE for VO2max was 3.41±022 L/min (60.3 ± 1.4ml O2/kg/min). There was a significant linear relationship between normalized VO2 and normalized TSI (intercept + βx; 0.120 + 0.889x, p<0.001); O2Hb (-0.937+ 1.918x, p<0.001); and HHb (0.290 + 0.724x, p=0.010). CONCLUSIONS: Excellent intrasession reliability of all NIRS parameters evaluated indicates changes across multiple running speeds can be attributed to physiological mechanisms, rather than measurement error. The observed relationships between VO2 and NIRS parameters pave way for future studies targeting altered running conditions such as speed, gradient, and gravity simulations." @default.
