- W2463416615 abstract "The extravasation of leukocytes out of the blood stream is the fundament of many physiological functions of the immune system as well as a main element of pathophysiological processes [1]. Aim of this study was to establish a microscopic approach that allows for the in vivo investigation of the cellular and molecular mechanisms of the whole emigration process in mice. Leukocyte rolling, adhesion, and transendothelial migration were investigated in the cremaster muscle C57BL/6 mice under control as well as under inflammatory conditions after local application of either PAF (100 nM, platelet activating factor) or KC (6,25 nM, murine chemokine acting on CXCR2) [2]. Leukocyte-endothelial cell interactions (rolling and adhesion) were analyzed conventionally after labeling of leukocytes with rhodamine 6G in vivo. Transmigration was observed and quantified with near-infrared reflective transillumination. This microscopic method provides a pseudo-three-dimensional plasticity of cellular structures. Stimulation with either PAF or KC decreased the number of rolling leukocytes (n/30 sec) significantly (p < 0.05) after 30min from 17.4 ± 2.6 (control) to 4.8 ± 0.5 (PAF) and 9.8 ± 2.8 (KC). In contrast, the number of adherent leukocytes (n/104 μm2) was significantly increased from 3.6 ± 0.9 to 18.6 ± 1.3 and 9.6 ± 1.8, respectively. After 60 min, this was followed by a significant increase in the number of emigrated leukocytes (n/104 μm2) from 5.0 ± 1.6 to 22.5 ± 2.3 and 13 ± 2.2, respectively. Taken together, we present a novel microscopic approach featuring the combination of conventional fluorescence microscopy and near-infrared reflective transillumination. This method allows for the in vivo investigation of the dynamic process of leukocyte emigration and the underlying cellular and molecular mechanisms in mice." @default.
