- W2466718583 abstract "Allograft rejection is triggered by T lymphocytes of the recipient that recognize donor MHC molecules. Clinically used immunosuppressive protocols suppress the immune system in the recipient non-specifically. On the long term, the goal must be to suppress the immune response against an allograft in a more specific manner. The aim of this study was to determine T cell mediated effects after immunization of the recipient with synthetic class II MHC allopeptides in a kidney and small bowel transplant model. The results of this study demonstrate that T cells were activated by the synthetic donor specific class II MHC allopeptide RT1.Duβ (20 – 44) via the indirect pathway of allorecognition, triggering a specific immune response against the allograft. Immunization with this peptide caused an accelerated rejection after kidney or small bowel transplantation, both after preoperative and postoperative immunization with a Th1 type cytokine profile (IL-2 and IFN-γ). The data suggest that the indirect pathway of allorecognition may play an important role in acute rejection. Therefore, synthetic peptides in combination with an immunsuppressive protocol could be used not only to specifically prevent allograft rejection, but to manipulate the immune system in order to induce tolerance." @default.
