- W2466787081 abstract "Certain individuals (here referred to as Non-Demented with AD neuropathology, NDAN) remain cognitively intact despite presenting with neuropathology (plaques and tangles) that is normally associated with symptomatic stages of Alzheimer's Disease (AD). This suggests that these individuals are naturally resistant to the cognitive onslaught of AD. Thus, understanding the molecular mechanisms involved in such resistance may provide novel, effective therapeutic targets for AD-related cognitive decline. We have previously shown that NDAN have preserved synaptic function and increased neurogenesis as compared to AD, but the underlying mechanisms remained unresolved. In order to address this important question, in the present work we measured the CNS levels and epigenetic regulation of BDNF, a neurotrophin involved in both synaptic health and sustained neurogenesis. Western immunoblots, ELISA, real time PCR (RT-PCR) and DNA methylation assays were used in hippocampus and frontal cortex from control, AD patients and NDAN subjects. In comparison to control, BDNF levels were reduced in the hippocampus and cortex of AD subjects, as determined by Western blot and quantified by ELISA. On the other hand, NDAN individuals showed significantly higher BDNF as compared to AD patients, however still lower than control. Quantification of BDNF mRNA by RT-PCR confirmed these results, with NDAN subjects showing a much milder reduction than AD patients when compared to control subjects. DNA methylation (an index of epigenetic suppression of gene expression) was observed in the BDNF promoter region from both AD and NDAN samples, however to a much lesser extent in these latter. These data suggest that the epigenetic down-regulation of BDNF mRNA and protein expression observed in the hippocampus and cortex of AD patients is attenuated in NDAN subjects, thus leading to overall higher tissue levels of BDNF. Given the role played by BDNF in promoting synaptic plasticity and sustained neurogenesis (both phenomena observed in the NDAN brain) our results suggest that sustained BDNF expression is an important factor in NDAN ability to preserve cognitive integrity despite the deposition of plaques and tangles.
