- W2475146539 abstract "There is a large diversity of oligosaccharide sequences linked to proteins. The molecules classically called glycoproteins comprise serum and cell membrane glycoproteins of approximate mol-wt range 20–200 kDa, having oligosaccharide chains linked to Ser/Thr or Asn, i.e., O- and N-linked, respectively, making up between 10 and 60% by weight. The structures of these chains are introduced in Chapters 2 – 12 of this book and the variety of glycosylation assessed in the Glossary. Additionally covered are (1) mucins, which are traditionally defined as larger mol-wt glycoproteins of 106 kDa and upwards having >60% oligosaccharide, mainly O-linked via GalNAc-containing oligosaccharide cores, and (2) proteoglycans or glycosaminoglycans, which also have a high carbohydrate/protein ratio, but classically have disaccharide repeating units with an alternate uronic acid residue and a large degree of sulfation. The distinction between these categories is becoming increasingly blurred, and they can now be seen as a spectrum of the aforementioned glycosylation patterns occurring on high- and low-mol-wt, secreted, and cell-surface glycoproteins. As examples of this diversity, classical mucin and proteoglycan sequences can occur on cell-membrane-attached proteins of relatively low mol-wt (1); glycoproteins and proteoglycans are found in forms attached to the membrane by lipid-linked anchors (ref. 1 and Chapter 8 ), and cytoplasmic glycoproteins having GlcNAc linked to Ser and Thr (ref. 2 and Chapter 14 ) are now also known. Additional oligosaccharide-to-protein linkages are found in proteoglycans (1) and bacterial glycoproteins (ref. 3 and Chapter 9 ).
