The Bedlington Terrier breed of dog has been shown to be afflicted with a recessive genetic trait that results in a reduced excretion of copper from the liver. Chronic active hepatitis and cirrhosis of liver are common among Doberman Pinscher breed of dogs8. Dogs that are homozygous for the copper toxicosis genes cannot tolerate normal dietary levels of copper. High levels of copper have been documented in some dogs with cirrhosis3. The cause or effect relationship between cirrhosis and increased hepatic copper is unclear except among the Bedlington Terrier dog. Hence, the study was undertaken to know the relationship of copper concentration in liver with that of the histological changes in the liver diseases. Thirty liver samples were collected from mature dogs of both sexes belonging to many breeds showing gross changes during routine post mortem examination to study histological and histochemical changes and atomic absorption analysis. Paraffin sections (5μm thickness) were stained with haematoxylin and eosin1. Duplicate sections of all the 30 liver tissues were stained with modified Rhodanine staining technique to demonstrate the copper and copper associated protein, which stained copper red to orange red, nuclei blue and bile in green colour1. Positive control tissues of liver for the copper histochemical staining were obtained from the Bedlington Terrier dog from USA. Hepatic copper concentrations were determined from dry ashed liver samples by atomic absorption spectroscopy6. Liver tissues of 0.5 cm or less in diameter were dried to constant weight at 100°C for 24 hours in hot air oven. Dry weight of liver tissues was recorded. Then the liver tissues were ashed in a muffle furnace at 450°C for 3 hours. The ash obtained was dissolved in 1.0 N HCl and analyzed in Perkin Elmer Model 3110 Atomic Absorption Spectrometer. All results were expressed as parts per million (ppm) parts on dry weight basis. The gross lesions observed were pale, enlarged liver with rounded borders in a few cases and mostly the liver was nodular having irregular borders, shrunken and hard on palpation. Cirrhosis of liver was observed in five cases.
