- W2479456987 abstract "Systems biology has been central to Caenorhabditis elegans research from the first steps of establishing the worm as a major model system through to the post-genomics era. Concepts such as completeness have been central to the philosophy of the worm community from its inception, and the standard systems approach to analyzing biology has been hard-wired into C. elegans research: first find all the components, then map all their connections, and finally examine how the network-level properties of the system explain the biological behavior of the organism. In this chapter, we first outline the initial phase of worm research from the seminal achievements in the worm in the pre-genomics era to the completion of the full genome sequence in 1998. At that point, both the anatomical phenotype and the genotype of the worm were completely known, and the central part of this chapter will discuss the various systems level approaches to understand how the instructions encoded in the 97Mb genome are interpreted to generate the 959 cells of the adult worm. This includes separate sections on genetic screens using both mutagens and RNA-mediated interference, RNA expression and gene regulation, phenotypic variation among isogenic individuals, proteomics approaches, and a section illustrating how computational approaches can integrate diverse large-scale datasets to uncover the molecular machineries that underpin worm biology. Finally, we highlight approaches that have received less attention from the C. elegans community, but that hold some promise as part of the quest to achieve a ‘complete’ understanding this tractable organism. These include dynamic and mechanistic modeling, the interplay between mechanical and biochemical processes, and the linking of natural genetic and phenotypic variation." @default.
