It allows for immediate cellular responses to physiological stimuli without the need for de novo mRNA synthesis, and is important for several critical cellular processes including growth, proliferation, and differentiation. mRNA translation, in particular initiation, is tightly regulated by a number of mechanisms. This chapter describes the signaling pathways that mediate these phosphorylation events, their effect on initiation factor activity, and their significance in the regulation of translation initiation. It also discusses the role of mRNA translation in cancer. The process of translation is divided into three stages: initiation, elongation, and termination. Translation initiation is a highly ordered process that is regulated primarily by phosphorylation of initiation factors, in particular those that are involved in 5' mRNA cap recognition and eIF4F complex formation. Increased expression or differential phosphorylation of these initiation factors leads to changes in cellular translation rates, which can result in drastic changes in growth, proliferation, differentiation, and survival. Due to its role in cellular growth and proliferation, the control of mRNA translation is implicated in the development of cancer. Therefore, eukaryotic cells have evolved intricate mechanisms to regulate mRNA translation initiation. Despite the recent success of anticancer therapies designed to inhibit translation, either by targeting eIF4E directly or suppressing mTOR, it is imperative to continue to study the mechanisms of translational control in order to fully understand the process of translation initiation and its role in human disease." @default.
