In a study described in the chapter, two approaches were used to demonstrate that a number of peptide hormones present in the pituitary gland are initially synthesized as part of a large common precursor. The peptide approach showed that in the cell line AtT-20-D16v, a newly synthesized polypeptide can be immunoprecipitated with either ACTH or β-endorphin antibodies. Another approach took advantage of in vitro mRNA translation. Total mRNA was extracted from AtT-20-D16v cells and translated in an mRNA-dependent protein-synthesizing cell-free system. Following immunoprecipitation with anti-ACTH and anti-β-endorphin antibodies, it was demonstrated that it is possible to find a common precursor of 28,500 D containing the sequences of ACTH and β-endorphin, as shown by tryptic and chymotryptic analyses. The full sequence of this precursor was obtained by cDNA cloning and nucleic acid sequencing and shows that the bovine ACTH/endorphin precursor is 239 amino acids long. This precursor contains a transitory signal sequence 26 amino acids long and an amino-terminal part 103 amino acids long containing the sequence of a melanocyte-stimulating hormone, γ-MSH." @default.
