- W2489721737 abstract "This chapter highlights some reasons different hit to lead (HtL) strategies are adopted in the rapidly changing climate of drug discovery. It presents a representative HtL model, which illustrates the process of hit list profiling, identification of key questions, HtL strategies, and series selection. The goal of the HtL campaign should be the discovery of new and promising leads by reducing the vast amount of chemical space in the most efficient way possible. A way to narrow down this chemical space and make educated designs is the establishment of an emerging pharmacophore model, and then to design molecules that probe the electronics and topology of the model. One way to sample chemical space is through matched pair analysis. In contrast to a matched pair strategy, statistical molecular design attempts to provide information-rich libraries with the minimal number of representative compounds based on statistical modeling." @default.
