- W249181630 abstract "Modern therapeutics for cancer are, in general, based on the mechanism-based selective inhibition of kinases or inactivation of deoxyribonucleic acid (DNA) polymerases to inhibit cancer cell proliferation. gem-Difluorinated nucleoside analogs, such as gemcitabine, are mimetics of their nonfluorinated nucleosides, and thereby competitively bind to the DNA polymerase and suppress chain elongation. Afatinib, a recently Food and Drug Administration (FDA)-approved pharmaceutical for first-line treatment of metastatic non–small cell lung cancer, is a mechanism-based inhibitor of epidermal growth factor receptor (EGFR) tyrosine kinases, and is also a potent inhibitor of the drug-resistant versions of EGFR tyrosine kinases. Fluorinated taxoid derivatives conjugated to vitamins, or immunoglobulin G monoclonal antibodies, are designed for cancer-cell-specific drug delivery. Biotin conjugates of taxoids with an 18F-labeled alkyl side chain serve as positron emission tomography tracers for monitoring the biodistribution of these drug conjugates. This chapter outlines synthesis and mechanisms of action of various fluorinated anticancer agents that include receptor tyrosine kinase inhibitors, fluorinated versions of taxoids, and the recently FDA-approved anticancer therapeutics such as ponatinib, clofarabine, afatinib, sorafenib, regorafenib, trametinib, dabrafenib, and vemurafenib." @default.
