• W2495115834 abstract "Impairment of septohippocampal cholinergic system plays an essential role in memory disorders. Function of this system may be modulated by challenge of a centrally active muscarinic antagonist scopolamine and an acetylcholinesterase inhibitor donepezil. Their modulatory effect in animals has been extensively studied using a Morris Water Maze (MWM) task which engages allocentric (world-centered) and egocentric (self-centered) spatial navigation. The present study investigated the effect of scopolamine and donepezil on different types of spatial navigation in humans using a human analogue of the MWM task. Twenty-three cognitively healthy young volunteers (M=22.1, SD=2.2 years) were administered scopolamine (0.6 mg), scopolamine (0.6 mg) with donepezil (5 mg) and placebo using a double-blind 3-way crossover design. Egocentric, allocentric and combined (egocentric and allocentric) spatial navigation tasks were assessed in the real-space human analogue of the MWM before and 2, 4 and 8 hours after drug administration. Memory status was determined by Rey Auditory Verbal Learning Test. Repeated measures general linear models were used in the analyses. Scopolamine significantly impaired performance on the combined task 2 and 8 hours after drug administration compared with placebo (p=.006, Cohen's d=0.56 and p=.019, Cohen's d=0.50) and performance on the allocentric task 4 hours after drug administration compared with combination of scopolamine and donepezil (p=.017, Cohen's d=0.51). Scopolamine did not affect performance on the egocentric task (p≥.571, Cohen's d≤0.18). The study extends findings from animal research of a scopolamine-induced allocentric spatial navigation deficit to humans. The use of a human analogue of the MWM in future pharmacological studies has potential to bridge preclinical and phase II/III data. Translational aspect of human analogue of the MWM reduces biases often found in memory tests in humans." @default.
