Nuclear magnetic resonance (NMR) spectroscopy is probably the most versatile technology for affinity screening in a low to medium throughput format due to a variety of possible assay configurations that offer different levels of information content. The application of NMR in the hit-to-lead phase is very much dominated by its use in fragment screening, which can be primarily used to generate leads and can also complement other approaches to assess. SPR and optical waveguide grating (OWG) are truly label-free methods and enable the detection of small changes in mass upon inhibitor binding to protein targets that are typically tethered to the biosensor. Isothermal titration calorimetry has the application of thermodynamic data in the lead generation process.
