- W2501807227 abstract "Despite huge investments in the pharmaceutical industry and advances in biotechnology and molecular biology, there has still been a lack of productivity increase in drug discovery over the last few decades, perhaps as a result of the extra development hurdles placed in the path of pharmaceutical research programs and the reduction in the number of tractable targets. This “productivity crisis” has triggered a lot of retrospective statistical analysis to understand what has possibly gone wrong and how to remediate problems in the drug-discovery process. Finding out the relationship between compound properties, quality, and drug clinical attrition has been a main focus of these kind of investigations. The underlying rationale is that filtering out compounds that are unlikely to be successful earlier in the drug-discovery process will largely save cost and resources. This chapter presents some advances on the studies of the ADMET (Absorption, Distribution, Metabolism, Excretion, and Toxicology) properties of compounds, promiscuity, drug-likeness, and binding efficiency metrics. Some interesting learning points from these studies are also discussed." @default.
