- W2502161010 abstract "Combinatorial peptide libraries with their extreme diversity are a source of ligands with properties to interact with proteins in a specific manner. In this chapter peptide libraries are presented as a mine whereby specific affinity ligands can be sorted out for chromatographic applications with the objective of isolating proteins of interest. Practical examples of screening and of protein purifications are discussed. As a second application, when a protein of interest is almost pure (e.g., more than 95% pure) and requires just a polishing operation to achieve a purity close to 100%, peptide libraries can also be used to advantage. Similarly to the dynamic concentration range reduction mechanism, a polishing operation is made by a simple loading but below the saturation level. Thus the main protein is recovered in the flow-through at a much higher purity compared to the initial loaded product. A third application that has great potential for development will also be discussed; this is the investigation of protein–protein interaction where the immobilized peptide library represents the deposit of epitope models that are the basis of the natural biological recognition. The potential use of peptide libraries will also be suggested within the domain of protein chemistry." @default.
