- W2502847799 abstract "This chapter discusses various aspects of display and selection of proteins on genetic packages (DSPGP). With the development of phage display, it is possible to obtain peptidyl compounds from as few as three or four amino acids up to proteins of several hundred amino acids that bind to almost any chosen target with affinities that range from picomolar (pM) up to micromolar (μM). DSPGP and phage display involve the construction and expression of vast collections of potentially useful proteins and selection of those mutated genes that specify novel proteins with actual desirable binding properties. The fundamental principle of the DSPGP method is one of forced or directed evolution. DSPGP selects for mutated genes that specify novel proteins with desirable binding properties by arranging that the product of each mutated gene be displayed on the outer surface of a replicable Genetic Package. Protein binding usually involves exclusion of bulk water in the interface between the protein and its binding mate. It is suggested that by using small proteins as parentals, one can obtain binding proteins that have the residues responsible for binding highly localized." @default.
