- W250739879 abstract "Differential scanning calorimetry (DSC) is a technique used to measure thermodynamics of solid- or liquid-phase transitions that produce or absorb heat. DSC is commonly applied in the biopharmaceutical setting for the characterization and engineering of manufacturable drug products. Proteins and peptides represent the largest sector of “Biologic” pharmaceuticals and this chapter describes the use of DSC for the thermal analyses of therapeutic proteins in dilute solutions and to a lesser extent in solid states. DSC measurements of solutions and solids are quite different and typically use separate instrument designs. Solution measurements require high sensitivity and are made using DSC instruments that are commonly classified as “microcalorimeters”, while solid-phase DSC is performed using “standard” DSC instrument designs. In this chapter, we describe the fundamental information DSC provides for protein therapeutics, a description of the instruments and parameters of their use, and finally common DSC applications within the biopharmaceutical setting." @default.
