This present study was carried out to investigate the effects of high fat diet (HFD) supplementation on brain oxidative stress, particularly in frontal cortical and hippocampal region of adult Sprague-Dawley rats. With regard to its neuroprotective potential, germinated brown rice (GBR) extract was used as an intervention (100 mg/kg and 200 mg/kg body weight). Rats were fed with HFD and also in combination with GBR extract for 24 weeks. Food intake, body weight changes, serum lipid profile, serum antioxidant status and glucose level were determined. Moreover, the mRNA expression of acetylcholinesterase (AChE), antioxidant and inflammatory-related biomarkers in frontal cortex and hippocampus were investigated using multiplex gene expression analysis. AChE level in the hippocampal region was also quantified. This study indicated that rats supplemented with GBR extract exhibited reduced weight, better lipid profile outcome, improved serum antioxidant and glucose level as well as regulates the expression of antioxidant and inflammatory-related genes in comparison to the untreated HFD group. Also, GBR extract was shown to reduce mRNA expression and the enzyme level of AChE in the hippocampus. In conclusion, this study highlights the possible mechanistic basis of GBR neuroprotective action in the brain, with potential implication on neurodegenerative disorders associated with oxidative stress resulted from high cholesterol consumption.
