Few studies are available in patients with progressive supranuclear palsy (PSP), while no study has been performed in patients with corticobasal syndrome (CBS). The purpose of this study was to investigate within- and between-network rsFC abnormalities in these two forms of taupathies, using independent component analysis. For this purpose we recruited eleven patients with PSP, 11 patients with CBS and 11 age and gender matched healthy subjects. Compared to healthy subjects, significant increase in within-network rsFC was observed in both PSP and CBS patients with more widespread abnormalities being present in PSP. Increased rsFC within the cerebellar network was positively correlated with motor scores. With respect to healthy subjects, functional connections between the salience network and executive control network as well as between the anterior and posterior components of default mode network were significantly decreased in PSP, those between the cerebellum and orbitofrontal networks were significantly increased in both PSP and CBS patients; rsFC between the orbitofrontal and sensorimotor networks was lower in PSP than in CBS. We also found between-network rsFC in salience network and executive control network positively correlated with Mini-Mental State Examination (MMSE) and negatively with motor severity in patients with PSP. The study provides evidence of functional brain reorganisation in both forms of taupathies, with more severe rsFC abnormalities in PSP. Within-network rsFC increase may represent low-frequency higher synchronisation in damaged neurons while between-network rsFC abnormalities may reflect loss of structural integrity of long–range white matter fibers.
