- W2516440312 abstract "Objective: To explore neurons with β oscillatory activity in the subthalamic nucleus (STN) in relation to parkinsonian symptoms. Background: Abnormal oscillatory activity in the basal ganglia has been predicted to play an important role in parkinsonian symptoms. Methods: 30 patients with Parkinson’s disease (PD) who underwent electrode implantation for STN DBS were studied. 13 patients were classified as akinetic-rigid (AR) type, 8 patients were classified as tremor dominant (TD) type and 9 patients were classified as mixed type. Microelectrode recording in the STN and the EMG on the limbs was simultaneously recorded. Single-unit and spectral analyses were performed. Coherence analysis was used. Unpaired t-test and chi-square were used to compare the differences among the three PD types. Results: Of 145 neurons identified in the AR type, 39.3[percnt] were β oscillatory neurons (mean: 20.3 ± 5.85 Hz, βFB), 0.7[percnt] were tremor frequency oscillatory neurons (4-6 Hz, TFB). Of 82 neurons identified in the TD type, 10.9[percnt] were β oscillatory neurons, 32.9[percnt] were TFB oscillatory neurons. Of 89 neurons identified in the mixed type, 23.6[percnt] were β oscillatory neurons, 19.1 [percnt] were TFB oscillatory neurons. There were significant differences in proportion of neurons with βFB and TFB oscillations among the three PD groups. There were TFB oscillatory neurons coherent with EMG activity of limb tremor in the TD type (n=17) and mixed type (n=7). Particularly, there were βFB oscillatory neurons coherent with limb EMG activity in the AR type (n=15), but there was no coherence in the TD type and mixed type. Conclusion: The STN βFB oscillatory neurons correlate with parkinsonian rigidity-bradykinesia. The high proportion of βFB oscillatory neurons found in the AR type is indirect evidence for their importance in generating motor symptoms. The quantity of pattern of oscillatory neurons seems to play a role in generation different type PD. Disclosure: Dr. Zhuang has nothing to disclose. Dr. Hallett has nothing to disclose.
