- W2516536450 abstract "Characterization of ribonucleic acid con- tent of particles released from cultures of marrow cells of leukemic patients indicates the presence of RNA molecules of size and base sequence characteristic of oncornarviruses. Seventeen marrow samples obtained from leukemic pa- tients in relapse or in a chronic phase of the disease yielded particles containing high-molecular-weight RNA with a sedimentation velocity (about 70 S) similar to that ob- tained for murine oncornavirus RNA. Eight of nine mar- row samples from non-leukemic patients did not yield de- tectable high-molecular-weight RNA. Among patients in firm hematological remission, three of three samples from patients with acute lymphoblastic leukemia and three of nine samples from patients with acute myeloblastic leu. kemia were positive for high-molecular-weight RNA. The base sequence of the RNA in particles was character- ized by synthesizing complementary (3H)DNA in an endo- genous reaction and hybridizing to excess RNA from known oncornaviruses. Hybridization of 40-60% of input complementary DNA to simian sarcoma virus RNA was detected. No homology was detected with an avian oncor- navirus (Rous sarcoma virus) while an intermediate level of homology (10-30%) was detected in hybridization to murine sarcoma virus (Kirsten) and murine leukemia viruses (Rauscher, Moloney, and Gross). Evidence has been gathered by a number of laboratories suggesting the presence of particles with biochemical and morphological properties characteristic of oncornaviruses in tissue from patients with all forms of leukemia (1, 2). An enzyme with the characteristics of an RNA-dependent DNA polymerase (reverse transcriptase) has been isolated from leukocytes of patients with leukemia (3, 4). We have re- ported that an enzyme activity with the properties of a re- verse transcriptase could be detected in marrow cells from leukemic patients; further, the enzyme activity increased in cultures of marrow cells from leukemic patients both in re- lapse and in remission (5). The increase was associated with release into the culture medium of particles having reverse transcriptase activity and having densities, biochemical properties, and morphology similar to that of known oncorna- viruses or oncornavirus cores (6). In this paper we present the results of experiments de- signed to characterize the nucleic acid comnponent of these particles.
