- W2519071605 abstract "Objective: Elevated maternal testosterone levels have been associated with pathological pregnancies such as preeclampsia which cause fetal growth restriction and increase the risk of development of cardiometabolic disorders in adult life. The study was therefore designed to investigate the effect of elevated maternal testosterone on glucose regulation, atherogenic lipids and fetal outcome. Design and Method: Pregnant Wistar rats were treated (s.c.) with either olive oil or testosterone (0.5 mg/kg b.w.) between gestational days 15 and 19. Glucose tolerance and insulin resistance was estimated by oral glucose tolerance test (OGTT) and homeostatic model of assessment (HOMA-IR) respectively. Plasma insulin and testosterone were determined by enzyme-linked immunosorbent assay. Total cholesterol (TC), triglyceride (TG) and high density lipoprotein-cholesterol (HDL-C) were determined by standardized enzymatic colorimetric methods while TG/HDL-C and TC/HDL-C ratios were evaluated as atherogenic indices. The fetal weight, placenta weight, number of fetus as well as fetal resorption were evaluated as fetal outcome. Results: The results show that elevated maternal testosterone led to increase in fasting blood glucose, 1hr-postload glucose level, insulin resistance, TG/HDL-C, TC/HDL-C, number of fetal resorptions. On the other hand, elevated maternal testosterone led to decrease in TG, HDL-C, fetal weight, placenta weight and number of fetus. However, elevated maternal testosterone did not affect glucose tolerance, plasma insulin and TC levels. Conclusions: The results from this study demonstrate that elevation of maternal testosterone would cause glucose deregulation and atherogenic dyslipidemia which may, at least in part, be responsible for the poor fetal outcome." @default.
