We haveemployedpolarized fluorescence confocal microscopy tomeasuretheorientation ofeosin-5-maleimide covalently reacted withLys-430 ofhuman erythrocyte band3.Steady-state polarized fluorescence imagesshoweddistinct intensity patterns, whichwere fit toan orientation distribution oftheeosin absorption andemission dipoles relative tothemembranenormalaxis. Thisorientation was foundtobeunchanged bytrypsin treatment, whichcleaves band3 betweentheintegral membranedomainandthe cytoskeleton-attached domain. Thisresult suggests thatphosphorescence anisotropy changesobserved after trypsin treatment areduetoa rotational constraint changerather thana reorientation ofeosin. Bycoupling time-resolved prompt fluorescence anisotropy withconfocal microscopy, we calculated theexpected amplitudes ofthee-ISande-4Dt termsfrom theuniaxial rotational diffusion modelandfoundthatthee-4Dt termshoulddominatetheanisotropy decay.Delayed fluorescence andphosphorescence anisotropy decaysofcontrol andtrypsin-treated band3inghosts, analyzed as multiple uniaxially rotating populations using theamplitudes predicted byconfocal microscopy, were consistent withthree motional species withuniaxial correlation times ranging from7,usto1.4ms." @default.
