- W2532424633 abstract "Alzheimer’s disease (AD) is a late-onset neurodegenerative disease, characterized by memory loss, depression, and multiple cognitive impairments. The co-morbidity of depression and impaired cognition is eminent with or without Alzheimer’s disease. Selective serotonin reuptake inhibitors (SSRIs have been used in aging populations to reduce the symptoms of anxiety, depression, and agitation. Recent studies investigating the efficacy of SSRI as treatment of these symptoms in elderly populations revealed reduced levels of circulating Aβ in the cerebrospinal fluid, suggesting that SSRIs may be therapeutic drugs for psychiatric disorders known to affect patients. However, underlying protective mechanisms of SSRIs for AD neurons are not known. To understand beneficial effects of SSRIs and Neurosteroids on Cell energy metabolism and cell survival in Serotonin and Alzheimer’s neurons in in vitro cell model. Rat serotonin cells and SHSY5Y cells will be treated with SSRI (citalopram and escitalopram) and Neurosteroids (Androgen, estrogen and Progesterone) to study the cell energy metabolism and oxidative stress. XF seahorse analyzer will be used to understand cell stress and bioenergy metabolism. Real time RT PCR of Serotonin and AD related genes and mitochondrial function. Cell apoptosis will be measure by CBA Annexin IV Assay. Neurosteroids and SSRI reduces AD pathology in Alzheimer’s neurons and cell survival in serotonin neurons. This study will allow us to understand impact of increasing serotonin on Amyloid beta pathology further we would like to see the energy metabolism and mitochondrial stress in Serotonin and AD neurons (SHSY5Ycells). As neurosteroids and SSRI increase serotonin in cells and hopefully increase cell metabolism as well. Serotonin is a key neurotransmitter plays a large role in synaptic functions of AD neurons. Our ongoing studies indicate that SSRI, and Neurosteroids target serotonin and AD neurons and reduce cell stress which results in reduced depression in mentally depressed individuals. In the current study, we propose to test the therapeutic targets and efficacies in serotonergic cells in vitro and AD neurons. Our proposed study give valuable information about serotonin impacting bioenergy metabolism, cell survival and benefit in Alzheimer’s therapy." @default.
